Marucci, Americo A. (Upstate Medical Center, Syracuse, N.Y.). Mechanism of action of staphylococcal alpha-hemolysin. I. Some factors influencing the measurement of alpha-hemolysin. J. Bacteriol. 86:1182-1188. 1963.-A kinetic method for the accurate and reproducible measurement of the action of staphylococcal alpha-hemolysin on rabbit erythrocytes is described. The activity of the alpha-hemolysin depends upon the temperature used for measurement. At 37 C the hemolysin lyses cells faster, but it in turn is rather quickly inactivated. At 0 C there is no inactivation, but the rate of lysis is greatly decreased. There is no change in the activity with change in total reaction volume, providing that the concentrations of cells and hemolysin are kept constant. The fraction of rabbit red cells lysed by a given amount of hemolysin in a given time is constant and independent of the total number of cells in the reaction mixture.

The Characterization of Staphylococcal Toxins

The continuous flow paper electrophoretic methods used to isolate alpha hemolysin are presented. The purity of the alpha hemolysin preparation is demonstrated by double agar diffusion, immunoelectrophoretic, and ultracentrifugation techniques. Indirect evidence of purity is provided by the fact that rabbit immunized with purified alpha hemolysin produced detectable antibody only to alpha hemoly...

Staphylococcus aureus α-Hemolysin Activates the NLRP3-Inflammasome in Human and Mouse Monocytic Cells

Community Acquired Methicillin Resistant Staphylococcus aureus (CA-MRSA) causes severe necrotizing infections of the skin, soft tissues, and lungs. Staphylococcal alpha-hemolysin is an essential virulence factor in mouse models of CA-MRSA necrotizing pneumonia. S. aureus alpha-hemolysin has long been known to induce inflammatory signaling and cell death in host organisms, however the mechanism ...

Membrane Bound Monomer of Staphylococcal α-Hemolysin Induces Caspase Activation and Apoptotic Cell Death despite Initiation of Membrane Repair Pathway

BACKGROUND Wild type Staphylococcal alpha-hemolysin (alpha-HL) assembly on target mammalian cells usually results in necrotic form of cell death; however, caspase activation also occurs. The pathways of caspase activation due to binding/partial assembly by alpha-HL are unknown till date. RESULTS Cells treated with H35N (a mutant of alpha-HL that remains as membrane bound monomer), have been s...

Anti-alpha-hemolysin monoclonal antibodies mediate protection against Staphylococcus aureus pneumonia.

Staphylococcus aureus pneumonia is one of the most common invasive diseases caused by this human pathogen. S. aureus alpha-hemolysin, a pore-forming cytotoxin, is an essential virulence factor in the pathogenesis of pneumonia. Vaccine-based targeting of this toxin provides protection against lethal staphylococcal pneumonia in a murine model system, suggesting that a monoclonal antibody-based th...

A molecular modeling based screening for potential inhibitors to alpha hemolysin from Staphylococcus aureus

Staphylococcus aureus, a Gram-positive bacterium is pathogenic in nature. It is known that secreted toxins remain active after antibiotic treatment. The alpha hemolysin or alpha toxin damages cell membrane and induces apoptosis and degradation of DNA. The titer of alphahemolysin increases and causes hemostasis disturbances, thrombocytopenia, and pulmonary lesions during staphylococcal infection...
